1. Sample Application: A small drop of whole blood (often via a sterile lancet and collection stick) is placed into each of the designated wells (Anti-A, Anti-B, Anti-D, and a Control well).
2. Mixing/Adding Diluent: In some kits, the blood is gently stirred with a provided stick; in others (e.g., solid-phase/dot-filtration methods), a sample diluent/buffer solution is added to the wells.
3. Observation: The results are observed within the specified timeframe (e.g., 2-30 minutes). 

• Agglutination (Clumping): Indicates the presence of that specific antigen on the red blood cells.
• No Agglutination (Smooth Mixture): Indicates the absence of that specific antigen.
